In recent years, several reports on the observation of the luminal surface of aorta using scanning electron microscope have been reported, but any of them failed to present the fine structure of the endothelial cell. The authors made some improvements upon the routine methods of preparation of specimens for electron microscopic research, and could observe the fine structure of the endothelial surface of aorta in dogs. The findings obtained were as follows : 1) By the single usage of any one of the fixatives such as formaldehyde, ethanol, acetone, Zenker's solution, Carnoy's solution, glutaraldehyde, and osmic acid, the authors could never have a clear finding of the structure of the luminal surface
